I’ve never made a rope, but have made quite a few plugs using a hydraulic press. I’ve found a few days makes a noticeable difference. My best luck has been pressing for a few days, then sticking it in a jar for a few months and forgetting about it. After a year or so some magic happens.

That being said, if you are starting with good quality tobacco, your rope should be great without much age.

It's going to dry out and not be as tight. It is probably still as tight as you want it. This is just academic. But a couple approaches are like, I think @Knucklehead does it with surgical tubing. I wrap in plastic before using fiberglass tape. Since jute is breathable, I think drying a little is good because it prevents mold, but theoretically you could keep it in a bag or humidor. I would do so with caution.
